Drug Interactions of Selumetinib
Concomitant Medications That Are Contraindicated
Concomitant use with the following drugs is prohibited:
Strong metabolic enzyme inhibitors: e.g., clarithromycin, grapefruit juice, ketoconazole tablets
Moderate metabolic enzyme inhibitors: e.g., erythromycin, fluconazole, omeprazole
Enzyme inducer drugs: e.g., phenytoin, rifampin, carbamazepine, Folium Forsythiae
Special Precautions
Selumetinib contains vitamin E; avoid additional vitamin E supplementation during treatment.
For patients receiving concomitant anticoagulants or antiplatelet agents, increase the frequency of coagulation function monitoring.

Before initiating treatment, fully disclose all medications (including prescription drugs, dietary supplements, and herbal medicines) to your physician for evaluation of potential interaction risks.
Adverse Reactions of Selumetinib
Severe Adverse Reactions
Include abnormal diarrhea, anemia (manifesting as fatigue, dizziness, etc.), and persistent fever.
Common Adverse Reactions
Vomiting, rash, diarrhea, nausea, fatigue, dry skin, fever, acne-like rash, decreased serum albumin (which may cause edema), and paronychia.
Management Recommendations
These reactions may be related to individual constitution and dosage. Monitor your condition closely during treatment. Seek immediate medical attention if you experience:
Worsening rash
Severe diarrhea (more than 3 watery stools per day)
Other severe symptoms; measure blood pressure, heart rate, and respiratory rate before medical evaluation.
Circumstances Requiring Immediate Discontinuation of Selumetinib
When adverse reactions occur, physicians will take appropriate measures based on severity:
Mild reactions: Temporary interruption of treatment for observation
Moderate reactions: Dosage reduction with continued treatment
Severe reactions: Permanent discontinuation of the drug
