Serious side effects of Selumetinib
During treatment with Selumetinib, if any of the following side effects occur, please consult your doctor immediately:
1. Very common (≥10%): skin blistering, crusting, irritation, pruritus, or redness; blood in urine; dry or cracked skin; diarrhea; rapid heartbeat; skin itching, pain, redness, swelling, tenderness, or warmth; nail loosening; muscle or bone pain; nosebleeds; redness or pain around nails; swelling (edema).
2. Less common (<10%): blurred.
Milder side effects of Selumetinib
Some side effects of Selumetinib are milder and may resolve as your body adjusts to the medication. If the following side effects significantly affect your daily life, please consult your doctor:
Very common (≥10%): constipation; fever; hair loss or thinning; loss of appetite; nausea; abdominal pain; oral swelling or inflammation (stomatitis); unusual tiredness or weakness; vomiting.

1. Overall adverse events
Most common adverse reactions (incidence ≥40%): vomiting, rash (all types), abdominal pain, diarrhea, nausea, dry skin, fatigue, musculoskeletal pain, fever, acneiform rash, stomatitis, headache, paronychia, and pruritus.
2. Cardiovascular system
Very common (≥10%): decreased ejection fraction (up to 23%), sinus tachycardia (20%), elevated blood pressure and hypertension (<20%).
3. Skin and subcutaneous tissue
Very common (≥10%): rash of various types (up to 91%), dry skin (60%), acneiform rash (50%), paronychia (including nail infections) (48%), pruritus (46%), dermatitis (36%), hair changes (32%), skin infections (20%).
Other reported events: palmoplantar erythrodysesthesia syndrome.
Most common rashes: acneiform dermatitis (54%), maculopapular rash (39%), and eczema (28%).
Rash types include acneiform dermatitis, maculopapular rash, erythema, pustular rash, rash, urticaria, exfoliative rash, pruritic rash, erythematous rash, etc. Acneiform rash refers to acneiform dermatitis. Dermatitis includes atopic dermatitis, diaper dermatitis, eczema, seborrheic dermatitis, and skin irritation. Hair changes include alopecia and hair color changes. Skin infections include abscess, cellulitis, impetigo, and staphylococcal skin infection.
4. Gastrointestinal
Very common (≥10%): vomiting (82%), abdominal pain (76%), diarrhea (up to 77%), nausea (up to 73%), stomatitis (including mouth ulcers) (50%), constipation (34%), elevated lipase (32%), dry mouth (<20%).
Other reported events: perforation, colitis, intestinal obstruction, ileus.
5. Blood and lymphatic system
Very common (≥10%): decreased albumin (51%), decreased hemoglobin (41%), neutropenia (33%), lymphopenia (20%).
6. Hepatobiliary system
Very common (≥10%): elevated aspartate aminotransferase (AST) (41%), elevated alanine aminotransferase (ALT) (35%).
7. Metabolism and nutrition
Very common (≥10%): increased blood potassium (27%), decreased appetite (22%), weight gain (<20%).
8. Musculoskeletal and connective tissue
Very common (≥10%): elevated creatine phosphokinase (CPK) (up to 79%), musculoskeletal pain (including limb, back, and neck pain) (58%).
Other reported events: rhabdomyolysis.
9. Nervous system
Very common (≥10%): headache (48%).
10. Ocular
Very common (≥10%): visual impairment (including blurred vision, photophobia, cataract, increased intraocular pressure) (<20%).
Other uncommon events (0.1% to 1%) reported: retinal vein occlusion (RVO), retinal pigment epithelial detachment (RPED), central serous retinopathy (CSR).
11. General disorders and administration site conditions
Very common (≥10%): fatigue and malaise/asthenia (56%), fever (56%), generalized and localized edema (including peripheral edema) (20%), facial and periorbital edema (<20%).
12. Respiratory, thoracic and mediastinal
Very common (≥10%): epistaxis (28%), dyspnea (including exertional and at rest) (<20%).
13. Renal and urinary
Very common (≥10%): hematuria (22%), proteinuria (22%), acute kidney injury (<20%).
