Main Efficacy

Alectinib is indicated as monotherapy for the treatment of patients with anaplastic lymphoma kinase (ALK)-positive locally advanced or metastatic non-small cell lung cancer (NSCLC).

Dosage and Administration

Patient Selection

Alectinib should be used in medical institutions with experienced professionals and under the guidance of qualified specialists.Prior to treatment, ALK positivity must be confirmed by a fully validated diagnostic test.

Recommended Dosage

Alectinib hard capsules should be taken with food, swallowed whole, and must not be opened or dissolved.The recommended dose is 600 mg (4 × 150 mg capsules) orally twice daily (total daily dose 1200 mg).

Duration of Treatment

Treatment is recommended until disease progression or unacceptable toxicity occurs.

Missed or Delayed Doses

If a dose is missed, it should be taken unless the next scheduled dose is due in less than 6 hours.

If vomiting occurs after dosing, the next dose should be taken at the scheduled time.

Dose Modification

For adverse events, dose interruption, reduction, or discontinuation may be required.Dose reduction should be done in 150 mg decrements twice daily based on tolerability.Treatment should be permanently discontinued if the patient cannot tolerate 300 mg twice daily.

Contraindications

Alectinib is contraindicated in patients with known hypersensitivity to alectinib or any of the excipients.
