What are the available dosage forms and strengths of loratinib?
Loratinib is currently only available in tablet form, with two different strengths:
25mg specification: The tablets are light pink round film-coated tablets with the word "Pfizer" engraved on the surface and "25" and "LLN" markings engraved on the back. After removing the outer coating, the tablets are white.
100 mg specification: The tablets are dark pink oval film-coated tablets with the word "Pfizer" engraved on the surface and "LLN100" engraved on the back. After removing the outer coating, the tablets are also white.
It's important to note that the daily dosage needs to be 100mg, which means you need to take both strengths of the tablets together. For example, you can choose four 25mg tablets, or take one 100mg tablet directly.
What are the specific uses of loratinib?
The standard dosing regimen is to take 100mg once daily at a fixed time. It can be taken before or after meals, regardless of food intake. The tablet must be swallowed whole; do not chew, break, or split it. This is to ensure accurate dosage and proper release of the medication.
If adverse reactions occur, the dosage needs to be adjusted. The specific method for reducing the dosage is as follows:
First adjustment: 75mg once daily. For 25mg tablets, take 3 tablets; if using 100mg tablets, break the tablets before taking, but in practice, it is recommended to directly switch to the appropriate tablet strength.
Second adjustment: 50mg once daily, equivalent to two 25mg tablets. If this dose is still not tolerated, medication should be discontinued completely.
The entire course of medication needs to continue until the disease worsens (such as tumor enlargement or metastasis), or until adverse side effects that the body cannot tolerate occur. A doctor must assess the specific situation before each dose adjustment.
