Mechanism of Action
Baricitinib is an immunomodulatory drug classified as a Janus kinase (JAK) inhibitor. Its active ingredient suppresses excessive immune system activation, and it is primarily indicated for the treatment of adult patients with moderate-to-severe rheumatoid arthritis who have had an inadequate response to other antirheumatic medications.
Indications
Baricitinib is specifically approved for the treatment of moderate-to-severe active rheumatoid arthritis in adult patients, particularly those who have failed to achieve adequate efficacy or cannot tolerate side effects after treatment with one or more conventional antirheumatic drugs (e.g., methotrexate).
It can be used as monotherapy or in combination with conventional antirheumatic agents. Common clinical regimens include combination with methotrexate or other non-biologic antirheumatic drugs.
Symptom Relief
Baricitinib effectively improves multiple symptoms associated with rheumatoid arthritis:
Joint redness, swelling, and pain: Especially swelling and tenderness in small joints such as the fingers and wrists.
Limited joint mobility: Manifested as morning stiffness and difficulty with extension and flexion.
Joint deformity: Structural changes in joints caused by chronic inflammation, such as enlargement and deformity of finger joints.
Other Approved Uses
In addition to rheumatoid arthritis, baricitinib has the following approved applications:
1.Treatment of severe COVID-19: For adult hospitalized patients requiring oxygen therapy, mechanical ventilation, or extracorporeal membrane oxygenation (ECMO), to help control excessive immune responses.
2.Treatment of severe alopecia areata: For adult patients with extensive hair loss on the scalp, face, or body, characterized by sudden, widespread areas of hair loss.
Concomitant Medications
In the treatment of rheumatoid arthritis, baricitinib is commonly used in combination with the following medications:
Methotrexate: A foundational antirheumatic drug; combination therapy enhances therapeutic efficacy.
Other non-biologic antirheumatic drugs: Dosing regimens are selected based on individual patient circumstances.
Combination use requires physician assessment of the patient’s individual condition to appropriately design the treatment plan and dosage.
