Enasidenib: Uses, Mechanism of Action, and Indications

Enasidenib is an oral medication used to treat acute myeloid leukemia (AML) in adult patients. Acute myeloid leukemia (AML) is a type of cancer that affects the blood and bone marrow.

Enasidenib Indications

Enasidenib is indicated only for patients who carry a specific genetic mutation: the IDH2 (isocitrate dehydrogenase-2) mutation. Enasidenib is used to treat patients with AML that is relapsed (the disease has worsened again) following initial treatment, or that is refractory (unresponsive to treatment).

The safety and efficacy of Enasidenib in children have not yet been established.

Enasidenib must be used in conjunction with the RealTime IDH2 assay, a diagnostic tool capable of detecting specific IDH2 gene mutations in patients with AML. IDH2 gene mutations can cause immature blood cells to become cancerous and divide uncontrollably, thereby leading to AML.

Enasidenib Administration Guidelines

1. Take Enasidenib strictly as directed by your doctor. The recommended dosage is 100 mg taken orally once daily; it may be taken with or without food. The medication should be taken at the same time each day; do not split or crush the tablets.

2. If you vomit after taking the medication, miss a dose, or fail to take the dose at your regular scheduled time, take the missed dose as soon as possible on that same day, and then resume your regular dosing schedule the following day.

3. Patients should continue taking Enasidenib until the disease progresses or until intolerable side effects occur.

4. If certain side effects occur, it may be necessary to adjust the dosage or temporarily suspend treatment. It is generally recommended to continue treatment for at least 6 months, whenever possible. 

Enasidenib Specifications

Enasidenib is available in two oral tablet strengths: 50 mg and 100 mg. It is manufactured by Celgene.
