Tazemetostat Drug Interactions
1. Strong/Moderate CYP450 3A Inhibitors
(1) Strong inhibitors: Coadministration is contraindicated.
(2) Moderate inhibitors: Avoid concomitant use if possible; if coadministration is necessary, reduce the dose. After discontinuation of the inhibitor for 3 half-lives, the original dose may be resumed.
2. Dose Reduction Regimen When Coadministered with Moderate Inhibitors
(1) Current dose 800 mg twice daily → reduce to 400 mg twice daily.
(2) Current dose 600 mg twice daily → reduce to first dose 400 mg + second dose 200 mg.
(3) Current dose 400 mg twice daily → reduce to 200 mg twice daily.

Store at a temperature not exceeding 30°C (86°F).
Tazemetostat Monitoring
Oncologic monitoring: Secondary malignancies (e.g., acute myeloid leukemia [AML], myelodysplastic syndrome [MDS], T-lymphoblastic lymphoma [T-LBL]).
Important Reminders
1. Dietary precautions: Avoid St. John's wort, grapefruit, and grapefruit juice.
2. Risk of secondary malignancies: Inform your doctor immediately if you experience fatigue, easy bruising, fever, bone pain, or pale skin.
3. Male patients: Effective contraception should be used by partners during treatment and for 3 months after the last dose.
4. Females of childbearing potential: Use effective non-hormonal contraception during treatment and for 6 months after the last dose; inform your doctor immediately if pregnancy is suspected or confirmed.
5. Breastfeeding: Avoid breastfeeding during treatment and for 1 week after the last dose.
