The approved trade name in Laos is MEKTODX, with Lao registration number: 09L1096/24.
Binimetinib (MEKTODX) is an ATP non-competitive reversible inhibitor that inhibits the kinase activity of mitogen-activated extracellular signal-regulated kinase 1 (MEK1) and MEK2. MEK proteins are upstream regulators of the extracellular signal-related kinase (ERK) pathway, which promotes cell proliferation. In melanoma and other cancers, this pathway is often activated by mutated forms of BRAF that activate MEK. Binimetinib (MEKTODX) inhibits BRAF-mediated activation of MEK and suppresses MEK kinase activity.
Indications
Binimetinib (MEKTODX) is a kinase inhibitor indicated for:
In combination with encorafenib, for the treatment of patients with unresectable or metastatic melanoma with a BRAF V600E or V600K mutation.
In combination with encorafenib, for the treatment of adult patients with metastatic non-small cell lung cancer (NSCLC) with a BRAF V600E mutation.
Dosage and Administration
1. Melanoma
The recommended dosage is 45 mg orally twice daily in combination with encorafenib. Binimetinib (MEKTODX) may be taken with or without food.
The recommended dosage for patients with moderate or severe hepatic impairment is 30 mg orally twice daily.
2. Non-Small Cell Lung Cancer
The recommended dosage is 45 mg orally twice daily in combination with encorafenib. Binimetinib (MEKTODX) may be taken with or without food.
Adverse Reactions
1. Melanoma
The most common adverse reactions (≥25%) with Binimetinib (MEKTODX) in combination with encorafenib were fatigue, nausea, diarrhea, vomiting, and abdominal pain.
2. Non-Small Cell Lung Cancer
The most common adverse reactions (≥25%) with Binimetinib (MEKTODX) in combination with encorafenib were fatigue, nausea, diarrhea, musculoskeletal pain, vomiting, abdominal pain, visual impairment, constipation, dyspnea, rash, and cough.
