Letermovir tablets are an antiviral medication with the active ingredient letermovir, specifically indicated for the prevention of complications caused by cytomegalovirus (CMV) infection in adult patients undergoing allogeneic hematopoietic stem cell transplantation (HSCT). CMV can cause severe complications in immunocompromised individuals.

The drug acts by blocking a key step in CMV replication: it inhibits the activity of the viral DNA terminase complex (comprising pUL51, pUL56, and pUL89 proteins), which functions as the "assembly machinery" for viral replication. Biochemical assays and electron microscopy have demonstrated that letermovir disrupts the normal assembly of the viral genome and interferes with the maturation of viral particles. Studies on drug-resistant viral strains confirm that the drug targets this critical enzyme complex.

Indications of Letermovir Tablets

Letermovir is primarily indicated for adult patients undergoing allogeneic HSCT who have tested seropositive for CMV (indicating prior CMV infection). It is used to prevent two potential post-transplant outcomes:

1.Reactivation of latent CMV leading to active infection

2.Organ damage and other diseases resulting from CMV infection
