Indications for Ripretinib (Augtyro):
1. Ripretinib (Augtyro) is used to treat non-small cell lung cancer in adults that has spread to the chest cavity or other parts of the body (metastatic).
2. Ripretinib (Augtyro) can also be used to treat metastatic tumors in adults and children aged 12 years and older that have spread to other parts of the body (metastatic), are unresectable, or are unresponsive to other treatments.
Side Effects of Ripretinib (Augtyro)
Ripretinib (Augtyro) may cause serious side effects. Call your doctor immediately if you experience any of the following:
Respiratory problems:
Sudden chest pain, wheezing, dry cough, shortness of breath.
General pain problems:
Sudden pain or difficulty moving in the hip, wrist, or back.
Nervous System Problems
Dizziness, vertigo (spinning sensation), mood changes, loss of balance or coordination, problems with thinking, memory, or attention, hallucinations, or sleep disturbances.
High Uric Acid Levels in the Blood
Joint problems, stomach pain, decreased or absent urination, nausea, vomiting, pink or brown urine.
Muscle Problems
Unexplained muscle pain, tenderness, weakness.
Liver Problems
Loss of appetite, nausea, vomiting, right upper quadrant pain, fatigue, itching, dark urine, clay-colored stools, jaundice (yellowing of the skin or eyes).
Common Side Effects
Dizziness, fatigue; altered taste, nausea; constipation; numbness or tingling in the arms or legs, loss of balance or coordination, difficulty walking; muscle weakness; shortness of breath; hallucinations, confusion, forgetfulness, problems with thinking or memory.
Allergic Reactions
Hives, difficulty breathing, swelling of the face, lips, tongue, or throat.
Repotrectinib (Augtyro) Dosage and Administration
I. Usual Dosage
Usual adult dosage for non-small cell lung cancer:
Initial dose: 160 mg orally once daily for 14 days.
Maintenance dose: 160 mg orally twice daily.
Duration of treatment: Until disease progression or unacceptable toxicity occurs.
II. Administration
1. Repotrectinib (Augtyro) can be taken with food or on an empty stomach, but the method of administration should be consistent each time.
2. Swallow the whole capsule with warm water. Do not crush, chew, break, or open the capsule. Do not take any damaged capsules.
3. Frequent medical examinations are required before and during treatment with repotrectinib (Augtyro). Depending on the results of these examinations, your next dose may be delayed, reduced, or permanently discontinued.
III. Missed Dosage or Vomiting
Missed Dosage Management
If you miss a dose of Repotrectinib (Augtyro), skip the missed dose and take the next dose at the usual time. Do not take double the dose at once.
Vomiting Management
If you vomit shortly after taking Repotrectinib (Augtyro), do not take the missed dose. Take the next dose as scheduled.
Precautions Before Taking Repotrectinib (Augtyro)
1. Inform your doctor about your medical history
Neurological disorders, gout, lung disease, liver or kidney disease.
2. Bone health
May increase the risk of fractures.
3. Contraception Requirements
May render hormonal contraceptives ineffective; barrier methods of contraception are required.
Women should take a pregnancy test before using this medication and use contraception during treatment and for two months after stopping the medication.
Male partners should use contraception for four months after stopping the medication.
4. Contraindications for Breastfeeding
Breastfeeding is prohibited during medication and for 10 days after discontinuation.
